Quantitative aspects of thymidine uptake into the acid-souble pool of normal and polyoma-transformed hamster cells.

چکیده
The uptake of thymidine into an acid-soluble pool in normal hamster embryo cells and hamster tumor cells transformed by polyoma virus was studied. It was found that tumor cells exhibited a 4-fold higher capacity than normal cells to incorporate thymidine as phosphorylated products into this pool. This was correlated with a 5to 10-fold higher activity of thymidine kinase in the tumor cell lines. The uptake process was strongly inhibited by the thymidine analog iododeoxyuridine, partially inhib ited by deoxyuridine, and not inhibited by uridine.

متن کاملSelection of morphologically normal cell lines from polyoma-transformed BHK21/13 hamster fibroblasts.
A selective method was devised for the isolation of "revertants" from polyoma-transformed sublines derived from BHK21/13 Syrian hamster fibroblasts. A hybrid, polyploid subline was obtained by growing together, in mixed culture in the presence of aminopterin, two variant BHK21/13 sublines lacking either inosinic acid pyrophosphorylase or thymidine kinase. Whereas these variant sublines were res...
متن کاملCharacteristics of the syrian hamster ribonucleic acid present in cells transformed by polyoma, simian virus 40, or adenovirus 12.
Ribonucleic acids of known specific activities were separately prepared from hamster cells transformed by polyoma, simian virus 40 (SV40), and adenovirus 12. When increasing amounts of these labeled ribonucleic acids were added to nitrocellulose filters containing immobilized (14)C-hamster deoxyribonucleic acid (DNA), about 5% of the DNA sites were saturated in each case. An additive saturation...
متن کاملAntigenic Differences between Normal and Polyoma Virus-transformed Hamster Cells. I. a Quantitative Study of the Cytotoxic Effect of Antisera.
Rabbit antisera against hamster embryo cells and hamster cells which had been transformed by polyoma virus were assayed by single cell technics. Antiserum killed cells in the presence or absence of complement, and the two mechanisms of inactiva tion appear to be different. Kinetics of cell-killing by antibody and complement are described. The transformed cells studied appear to have less surfac...
متن کاملThymidine phosphate pools and DNA synthesis after polyoma infection of mouse embryo cells.
We investigated conditions for the incorporation of thymidine-3H into pools of thymidine compounds and into DNA in mouse embryo cells in culture with the purpose of measuring changes in pool sizes and their possible correlation to the rate of DNA synthesis. Thin and dense cultures of embryo cells were grown in Petri dishes during six days. Thymidine triphosphate was the dominating thymidine com...
